Characterization of phototrophic microorganisms and description of new cyanobacteria isolated from the saline-alkaline crater-lake Dziani Dzaha (Mayotte, Indian Ocean) ArchiMer
Cellamare, M.; Duval, C.; Drelin, Y.; Djediat, C.; Touibi, N.; Agogue, H.; Leboulanger, C.; Ader, M.; Bernard, C.
The saline-alkaline crater-lake Dziani Dzaha (Mayotte, Indian Ocean) is dominated by the bloom-forming cyanobacterium Arthrospira. However, the rest of the phototrophic community remains underexplored because of their minute dimension or lower biomass. To characterize the phototrophic microorganisms living in this ecosystem considered as a modern analog of Precambrian environments, several strains were isolated from the water column and stromatolites, and analyzed using the polyphasic approach. Based on morphological, ultrastructural and molecular (16S rRNA gene, 18S rRNA gene, 16S-23S ITS region, cpcBA-IGS locus) methods, seven filamentous cyanobacteria and the prasinophyte Picocystis salinarum were identified. Two new genera and four new cyanobacteria...

Cyanobacterial populations that build 'kopara' microbial mats in Rangiroa, Tuamotu Archipelago, French Polynesia ArchiMer
Richert, Laurent; Golubic, Stjepko; Le Guedes, Roland; Herve, Annie; Payr, Claude.
Cyanobacterial populations, the primary producers and builders of 'kopara' microbial mats were studied in four selected ponds along the rim of the Rangiroa Atoll, French Polynesia using a polyphasic approach. Seven isolates were maintained in uni-cyanobacterial cultures, characterized morphotypically and phylogenetically by evaluating sequences of the 16S rRNA gene of about 620 base pairs in length. Cyanobacteria in natural populations were analyzed microscopically, characterized morphotypically, and compared with cultured strains. Three of the isolates were identified in the field samples: Lyngbya aestuarii, Johannesbaptistia pellucida and Chroococcus submarinus were present in the mats only as minor components, whereas the species of Schizothrix that...

Diversity of rhizobia isolated from nodules of indigenous tree legumes from the Brazilian dry forest Acta Agron. (Palmira)
Pereira de Lyra,Maria do Carmo Catanho; Santiago de Freitas,Ana Dolores; Bastos da Silva,Maria Luiza Ribeiro; de Vasconcelos Bezerra,Rosemberg; Silva,Vinicius dos Santos Gomes da; Ferreira da Silva,Aleksandro; Mergulhao,Adália Ca.valcanti do Espirito Santo; Dantas,Edilándia Farias; de Rosália e Silva Santos,Carolina Etienne.
Abstract The diversity of rhizobial isolates in tropical dry forests (TDF) has been studied due to the great importance of finding new species of bacteria capable of fixing nitrogen in tree legumes. In the Brazilian TDF (caatinga), Leguminosae is the most important family of plants, so the knowledge of microbial communities that make symbioses with native plants can help in the understanding of the interaction plant-microorganisms, as well as in the optimization of biological processes that can improve the cultivation system in an area so rich and understudied. In this study was determined the characteristics of rhizobia isolated from nodules of Mimosa tenuiflora (Willd.) Poir., Piptadenia stipulacea (Benth.) Ducke and Mimosa caesalpiniifolia Benth, grown...

Molecular and phenotypic characterization of Vibrio aestuarianus subsp francensis subsp nov., a pathogen of the oyster Crassostrea gigas ArchiMer
Garnier, Matthieu; Labreuche, Yannick; Nicolas, Jean-louis.
Eleven Vibrio isolates invading the hemolymph of live and moribund oysters (Crassostrea gigas) collected in the field and from a hatchery in France, were characterized by a polyphasic approach. Phylogenetic analysis of 16S rRNA, gyrB and toxR genes indicated high homogeneity between these strains and the Vibrio aestuarianus type strain (ATCC35048(T)), and confirmed previous 16S rRNA analysis. In contrast, DNA:DNA hybridization was from 61% to 100%, while phenotypic characters and virulence tests showed a large diversity between the strains. Nevertheless, several common characters allowed the isolates to be distinguished from the reference strain. On the basis of several distinct phenotypic characteristics, it is proposed to establish two subspecies within...

Prorocentrum rivalis sp nov (Dinophyceae) and its phylogenetic affinities inferred from analysis of a mixed morphological and LSU rRNA data set ArchiMer
Delmail, David; Labrousse, Pascal; Crassous, Philippe; Hourdin, Philippe; Guri, Mathieu; Botineau, Michel.
A new freshwater epiphytic Prorocentrum species, Prorocentrum rivalis, from the temperate region of the Haute-Vienne, France, is described. This species is the third freshwater species identified among approximately 60 marine Prorocentrum species. This new species is described using scanning electron microscope and phylogenetic analyses by a polyphasic approach (LSU rRNA sequences combined with 9 morphological characters). The phylogenetic analysis attests that P. rivalis is close to other planktonic freshwater species and the freshwater Prorocentrum clade is evolutionarily derived from an epiphytic freshwater prorocentroid ancestor. The unique marine species in the freshwater clade results from an ecophysiological reversion. P. rivalis differs from other...
